OPTOLANE’s Digital Real-Time PCR technology is designed to have the easiest way to check the amplification status and quantitative results among conventional digital PCR technologies commercialized in the market. After injecting the sample to be analyzed into the dedicated digital real-time PCR cartridge (Dr. PCR 20K), insert it into the instrument and amplify it. Hands on time only needs 2 minutes. It does not require a complicated process like conventional digital PCR equipment. LOAA can do all the fields of applications that digital PCR can do.

Everything can be digitized​

Digital Real-Time PCR, which has superior sensitivity performance compared to general real-time PCR quantitative analysis, is capable of absolute quantification without a standard curve. It guarantees high specificity and sensitivity as it is less affected by various PCR inhibitors. In particular, LOAA has the smallest hands-on time among commercially available digital PCR devices, so it can minimize the occurrence of result errors due to deviations between experimenters during the experiment. Therefore, we present a high reproducibility.

It can detect and quantify traces of copy number variation, and mutant genes of low incidence targets, as well as viruses. By putting one cell in each of 20,000 independent wells and amplifying it, the number of expressed cells can be quantified, and whether or not the gene is expressed and changes in the amount of expression can be detected.

LOAA has two fluorescence detection channels. Yellow dots in lower left side represents negatives for both channels. Green dots are for channel 1 positives and Red dots are for channel 2 positives. Dual positives are represented by blue dots. In the upper right side, there are some yellow dots which may be categorized as positives in current digital PCR system. We can clearly determine negatives by the shape of the real-time PCR curves.

Genetic mutations such as tumors can be easily identified using nucleic acids extracted from blood without a complicated digital PCR setup process in liquid biopsy research fields that require confirmation of a very small amount of gene profiling through a very small sample. How it works

LOAA (Lab on an array) uses a dedicated cartridge that integrates the latest CMOS photo sensor-based semiconductor technology as a third-generation digital real-time PCR device that performs ultra-precision, ultra-high speed, real-time, ultra-light, and digital PCR. It is an in-vitro diagnostic medical device that maximizes usability so that anyone can easily perform digital PCR without the demanding partitioning process.

LOAA Dr. PCR is a method of checking and counting the amplification of each individual well after the PCR Mixture for amplification is divided into 20,000 independent wells. In individual wells, amplified wells become positive wells and non- amplified wells become negative wells, and the total number of positive wells can be calculated using Poisson distributions to determine the result by the number of clones per sample microliter by calculating the amplified target DNA.

Dr. PCR cartridge is partitioned into 20163 wells. As a result, the PCR mixture to be analyzed can be dispensed into the cartridge and placed directly into the instrument for amplification. This eliminates the tricky partitioning process such as creating droplets. Using the dedicated buffer, you can check the quantitative value within 1 hour as quickly as possible. In the case of RNA targets, a dedicated buffer for One step RT is provided so that amplification can be performed directly without a separate cDNA synthesis process.
